Can there be a difference between using a mesh vs using a point cloud (scanned surface) when using the ICP register feature? I assume it is the same with the caveat that points may be sampled from the mesh(s) to do the calculations.

Yes, it's all about how the points are sampled. It will be probably more regular with a mesh (otherwise with a point cloud only a random number of points is chosen at each iteration).
