I have measured points from a total station in *.txt format (X,Y,Z).
Can I calculate the volume from this?

The only tool to compute volume from a point cloud is the '2.5D Volume computation' tool (https://www.cloudcompare.org/doc/wiki/i ... .5D_Volume)
But it's working under quite specific conditions, and you'll need a very dense point cloud (which doesn't seem to be the case here?)

Unfortunately, I don't have a thick cloud. It's a matter of targeting a heap of soil with a total station. There are approximately 40 points in a 20x20 meter area.

Than you could try to mesh it (probably with Delaunay 2.5D) and if it looks viable, then sample a lot of points over this mesh (with Edit > Mesh > Sample points).
